چکیده
The brown citrus aphid, Toxoptera citricida (Kirkaldy), is an important pest of Florida citriculture because it causes feeding damage to citrus and vectors citrus tristeza virus. Parasitoids recovered from brown citrus aphids in Florida include Lysiphlebus testaceipes (Cresson), Lipolexis scutellaris Mackauer, and Aphelinus gossypii Timberlake. Monitoring the levels of parasitism caused byeach species is difÞcult because theparasitoidsmust be rearedoutordissected fromtheaphidhosts. A simple and quick molecular approach was developed to detect and distinguish these parasitoids developing within the host aphid. Total genomicDNAwas extracted from the brown citrus aphid and eachof the threeparasitoids and the18S rRNAgeneof each specieswas ampliÞedbypolymerase chain reaction (PCR). The PCR products were sequenced to obtain complete gene sequences for each species. Thevariable regionsV2of the geneswereused todesign species-speciÞcprimers for detecting and differentiating the three parasitoids. The species-speciÞc PCR ampliÞcations discriminated the parasitoid DNAs from each other and from the host DNA. Detection of L. testaceipesDNAwithin the host aphid was possible in 8% of samples during the Þrst 2 h after parasitoid oviposition; in 66% of samples after 24 h; in 94% of samples after 48 h; and in 100% of samples after 72 h. The PCR approach described in this study provides earlier and more precise detection of parasitism and determination of species than rearing or dissection methods.

Molecular differentiation of sheep and cattle isolates of Fasciola hepatica using RAPD-PCR
Understanding genetic structure and status of genetic variation of Fasciola hepatica isolates from different hosts, has important implications on epidemiology and effective control of fasciolosis. Random amplified polymorphic DNA (RAPD-PCR) was used to study the genetic variation of F. hepatica in sheep and cattle. DNA was extracted from adult helminthes removed from livers of each infected ani...
متن کاملPOPULATION ECOLOGY Relationship of Soybean Aphid (Hemiptera: Aphididae) to Soybean Plant Nutrients, Landscape Structure, and Natural Enemies
In the north central United States, populations of the exotic soybean aphid, Aphis glycinesMatsumura (Hemiptera: Aphididae), are highly variable across space, complicating effective aphid management. In this study we examined relationships of plant nutrients, landscape structure, andnatural enemieswith soybeanaphid abundance across Iowa,Michigan,Minnesota, andWisconsin, representing the range o...
متن کاملMetapopulation structure of the specialized herbivore Macrosiphoniella tanacetaria (Homoptera, Aphididae).
We investigated population dynamics, genetic diversity and spatial structure in the aphid species Macrosiphoniella tanacetaria, a specialist herbivore feeding on tansy, Tanacetum vulgare. Tansy plants (genets) consist of many shoots (ramets), and genets are grouped in sites. Thus, aphids feeding on tansy can cluster at the level of ramets, genets and sites. We studied aphid population dynamics ...

The advent of molecular biology in general and the polymerase chain reaction in particular have greatly facilitated genomic analyses of microorganisms, provide enhanced capability to characterize and classify strains, and facilitate research to assess the genetic diversity of populations. The diversity of large populations can be assessed in a relatively efficient manner using rep-PCR-, AFLP-, ...
